General description

Methylene blue is bis(dimethylamino)phenothiazine hydrochloride (tetramethylthionine),
C16H18N3SCl. It is a major basic histological dye in pharmacology and microbiology. This heterocyclic aromatic chemical appears as a dark green solid powder, changing to blue in an aqueous solution. Methylene blue is tested for histological and and bacteriological staining, and as a constituent of Wright’s blood stain.

Application

Methylene blue is widely used in several routine staining applications including Mallory’s phloxine-methylene blue on paraffin sections of tissue fixed in Zenker’s fluid for nuclear staining and bacterial staining in milk. It is also suitable for manufacturing the polychrome methylene blue used in the preparation of Romanowsky stains.Methylene Blue hydrate has also been used:
  • in the preparation of methylene blue (MB) solution
  • to stain zymograms -to label the injection site in the tissue slices
  • to pre-treat mice via oral gavage to study its effects on noise-induced hearing loss
