General description

Special quality for the cleavage of fusion proteins.

Specificity

The coagulation factor Xa is a serine protease which recognizes the amino acid sequence Ile—Glu—Gly—Arg ↓ with a high degree of specificity. Cleavage of this sequence activates the natural substrate prothrombin to thrombin. This sequence can also be used as a restriction cleavage site for processing of recombinant fusion proteins. When a lack of specificity is due to steric hindrance, the introduction of an additional amino acid sequence can sometimes overcome the problem.

Application

Processing of fusion proteins at definite cleavage site. The restriction protease factor Xa is a serine protease which recognizes the amino acid sequence Ile — Glu — Gly — Arg ↓ with a high degree of specificity. This sequence may also be used as a restriction cleavage site for processing of recombinant fusion proteins.

Preparation Note

Working concentration: 1:10 to 1:200 (w/w)
A useable working concentration has a substrate ratio of 1:10 to 1:200 (w/w).

Note: The contents of a vial may be used for several simultaneous digests. When repeating the digest, a new vial should be used. Following this procedure ensures reproducibility and prevents possible contamination.
Storage conditions (working solution): A solution in redistilled water can be used for up to one week when stored at 4 °C.
An aqueous solution, stored in aliquots at -20 °C is stable for approx. one month.
Freezing aliquots increases enzyme stability in solution.
