General description

Streptavidin, a tetrameric protein, has been extensively used in purification and detection of biomolecules. The strong streptavidin-biotin bond can highly suited for attaching various biomolecules to one another or onto a solid support. The streptavidin-biotin complex displays high resistance to organic solvents, denaturants, detergents, and proteolytic enzymes. It can also withstand extremes of temperature and pH. However, one challenge faced by researchers using the streptavidin-biotin system is the target aggregation, which results from multivalent binding. It can cause cross-linking that can interfere with observations at the molecular level. These challenges can be overcome by use of monomeric streptavidin (mSA) that displays structural and binding properties that are suitable for multiple applications based on its small size and monovalent biotin binding. The monomeric form allows for association with biotinylated ligands without inducing aggregation that are observed with multivalent interactions. Monomeric form can also be genetically fused to another protein to form a biotin-binding tag. SAvPhire Monomeric Streptavidin is a is a high-affinity recombinant variant of streptavidin that minimizes the risk of aggregation, precipitation, and crosslinking in pre-incubation steps with biotinylated molecules and offers easier access to the desired targets. Unlike other non-engineered monomeric forms, SAvPhire Monomeric Streptavidin displays highly desirable dissociation kinetics (koff = 63 x 10-3 min-1 for non-engineered monomeric form and 2.1 x 10-3 min-1 for SAvPhire Monomeric Streptavidin) and longer t1/2 (11 min vs 329 min). Specificity

SAvPhire Monomeric Streptavidin expressed in E. coli and conjugated with Alpha Fluor 488 that is suitable for immunodetection of biotinylated proteins.

Evaluated by Immunocytochemistry in A431 cells.



Immunofluorescent analysis of A431 cells performed using a 1:1 mixture of biotinylated Anti-EGFR (1:1,000 dilution) and SAvPhire Monomeric Streptavidin Alpha Fluor 488 detected EGFR.
