Antibody specificity was examined in the rat amygdala, cortex, and hippocampus. Detection of primary antibody with ABC according to manufacturers directions. Low levels of glutaraldehyde (0.1%-0.3%) may be used in conjunction with paraformaldehyde for tissue perfusion fixation. Addition of intensifying reagents such as nickel ammonium sulfate to the chromogen solutions allows further dilution of the primary antibody to approximately 1:1000 or greater. For staining paraffin sections, heating in 10 mM citrate buffer is required (see application references). Antibody should be titrated for optimal results in individual systems.
