General description

SUN domain-containing protein 1 (UniProt Q9D666; also known as Protein unc-84 homolog A, Sad1/unc-84 protein-like 1) is encoded by the Sun1 (also known as Unc84a) gene (Gene ID 77053) in murine species. SUN1 is a component of the SUN-protein-containing complexes known as LINC (linker of nucleoskeleton and cytoskeleton), which link the nucleoskeleton and cytoskeleton structures together in a scaffold like fashion. SUN1 is required for interkinetic nuclear migration and nucleokinesis as well as nucleus-centrosome coupling during radial neuronal migration and glial migration in the cortex. SUN1 also is important during meiosis and is required for proper gametogenesis. It also participates in anchoring chromosome movement in the prophase of meiosis and in selective gene expression of coding and non-coding RNAs required for gametogenesis. Additionally, it has a vital role in the distribution of nuclear pore complexes on the nuclear membrane. Interestingly, LINC complexes are also critical for vital functions such as hearing.

Specificity

Clone 12.10F was raised against a fusion construct composed of murine SUN1 exon 6-, 8-, and 10-encoded regions that are present in murine spliced isoforms 1, 3, and 4 reported by UniProt (Q9D666). The epitope location among these three regions has not been mapped and reactivity toward murine spliced isoform 3 is expected only if the epitope resides within exon 1- or 4-encoded region.

Immunogen

GST-tagged recombinant fusion protein composed of murine SUN1 regions encoded by exons 6, 8, and 10.

Application

Immunocytochemistry Analysis: A 1:50 dilution from a representative lot detected SUN1 nuclear immunoreactivity in 4% paraformaldehyde-fixed, 0.3% Triton X-100-permeabilized C2C12 murine myoblasts.
