General description

Podocalyxin (UniProt: Q9R0M4; also known as Podocalyxin-like protein 1, PC, PCLP-1) is encoded by the Podxl (also known as Pclp1) gene (Gene ID: 27205) in murine species. Podocalyxin is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 22-404), a transmembrane domain (aa 405-425), and a cytoplasmic domain (aa 426-503). Its expression is observed in podocytes, liver cells, hematopoietic progenitor cells, vascular endothelial cells, and a subset of neurons. It is present as a monomer but can oligomerize upon association with the membrane raft. Its O-glycan-rich domain in the extracellular region and the C-terminus PDZ-binding motif in the cytoplasmic tail harbor an apical sorting signal. Its cytoplasmic domain is reported to be essential for the apical membrane targeting and renal tubulogenesis. Its large highly anionic extracellular domain allows to maintain open filtration pathways between neighboring podocyte foot processes. The extracellular domain is also required for microvillus formation. In kidneys, it functions primarily as an anti-adhesive molecule in podocyte epithelial cells, regulating adhesion and cell morphology. In other tissues, it plays subtle roles in tissue remodeling and development. It also participates in a number of cellular functions through its association with the actin cytoskeleton, ezrin, and Na+/H+ exchanger regulatory factor 1 and 2 proteins. It is the most commonly used marker protein for detecting podocytes in urine under conditions such as IgA nephropathy, Henoch-Schönlein purpura, diabetic nephropathy, lupus nephritis, poststreptococcal glomerulonephritis, membranous nephropathy, and focal segmental glomerulosclerosis. Higher expression of podocalyxin is observed in many cancers, especially those that originated from the epithelium and is linked to poor prognosis, more aggressive tumor progression, and chemoresistance.
