General description

Rho guanine nucleotide exchange factor 28 (UniProt P0C6P5; also known as 190 kDa guanine nucleotide exchange factor, p190-RhoGEF, p190RhoGEF, Rho guanine nucleotide exchange factor) is encoded by the Arhgef28 (also known as Rgnef) gene (Gene ID 361882) in rat species. p190RhoGEF is an ubiquitously expressed protein that belongs to the same subclass of Dbl family GEFs as Lbc, Lfc, GEFH-1, and p115RhoGEF. It contains several potential regulatory motifs, including an N-terminal leucine-rich region, a cysteine-rich zinc-finger domain and a C-terminal region that may form an alpha-helical coiled-coil. p190RhoGEF binds to RhoA, but not with Rac and Cdc42. When overexpressed in neuronal cells, it induces cell rounding and inhibits neurite outgrowth. Endogenous p190RhoGEF is reported to localize to distinct RhoA-containing regions at the plasma membrane, to the cytosol and along microtubules. In vitro and in vivo binding experiments show that p190RhoGEF directly interacts with microtubules via its C-terminal region adjacent to the catalytic Dbl homology/pleckstrin homology domain. Specificity

Clone 14C10.1 targets an epitope within the N-terminal portion of ARHGEF28 microtubles-interacting domain.

Immunogen

GST-tagged recombinant rat ARHGEF28 fragment encompassing the N-terminal portion of microtubles-interacting domain, excluding sequence C-terminal to the RNA-binding region.

Application

Detect ARHGEF28 using this mouse monoclonal Anti-ARHGEF28, clone 14C10.1, Cat. No. MABS1685, validated for use in Immunohistochemistry (Paraffin) and Western Blotting.
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected ARHGEF28 in rat heart and kidney tissue sections.

Quality

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ARHGEF28 in 10 µg of PC12 rat pheochromocytoma cell lysate.

Target description

~190 kDa observed. 190.8 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
