General description

G-protein coupled receptor kinase 2 (Grk2), also known as BARK or BARK1, is a member of the large family of GRK kinases that phosphorylate G-protein coupled receptors (GPCR), resulting in the recruitment of arrestins to the cytoplasmic domains of GPCR’s and subsequent internalization of these receptors by endocytosis. GRK2 is ubiquitously expressed, and modulates diverse cellular processes through association with a number of non-receptor substrates. GRK2 regulates actin dynamics and cell motililty through interaction with radixin, tubilin, and ezrin. In addition, Grk2 may play a role in the signaling of TGFβ by blocking the nuclear translocation of Smad2 and Smad3 proteins. It has also been shown that Grk2 may play a role in the hedgehog signaling pathway during embryonic development, by modulating the signaling of the atypical G-protein coupled receptor Smo. Grk2 is inactivated by phosphorylation of ser670 by ERK. It is also phosphorylated by PKC and Src, depending on the cellular context.

Application

Anti-phospho-GRK2 (Ser670) Antibody, clone 15G11.1 is an antibody against phospho-GRK2 (Ser670) for use in Western Blotting, Immunoprecipitation, Peptide Inhibition Assay.
Peptide Inhibition Assay Analysis: A 1:1,000 dilution from a representative lot blocked phospho-GRK2 in 10 µg of Calyculin A/Okadaic Acid treated and untreated Jurkat cells.

Immunoprecipitation Analysis: A representative lot immunoprecipitated phospho-GRK2 in HeLa cell lysate.
