Protein delta homolog 1or DLK-1 (DLK1), also known as pG2 and the Preadipocyte factor-1 (Pref-1), is a single pass type 1 membrane protein known to play a steering role in neuroendocrine differentiation. The protein is expressed within the stromal cells in contact with the vascular structure of the placental villi, yok sac, fetal liver, adrenal cortex and pancreas during development and DLK1 is also expressed within the beta cells of the islets of Langerhans in the adult. DLK1 plays an important gene regulatory expression role during the development of not only these very early structures but also in the regulation of the development of many specialized cells and tissues such as adipocytes. In adipose tissue, DLK1/Pref-1 is specifically expressed in preadipocytes but not in adipocytes and thus can be used as a preadipocyte marker. The inhibition operates through the creation of a soluble cleaved cytoplasmic form of DLK1 via the action of the metalloproteinase ADAM17 on mature DLK1. Fetal Antigen 1 (FA1) or soluble DLK-1/Pref-1 activates the MAPK kinase/ERK pathway, and via Sox9 interactions ultimately inhibits the differentiation of pre-adipocytes into mature adipocytes. Furthermore, by inducing Sox9, Pref-1 promotes chondrogenic induction of mesenchymal cells but prevents chondrocyte maturation as well as osteoblast differentiation. Thus, DLK1/Pref-1 directs multipotent mesenchymal cells (MSCs) toward the chondrogenic lineage but inhibits differentiation into adipocytes as well as osteoblasts and chondrocytes. Naturally, DLK1 has multiple isoforms each with slightly differing but related influences on MSCs development. Finally, DLK1 is also expressed in various tumors including small cell lung carcinoma and numerous neuroendocrine cancers.

Purified recombinant fragment of human DLK1 expressed in E. Coli.

Application

Detect DLK1 using this mouse monoclonal antibody, Anti-DLK1 Antibody, clone 3A10 validated for use in IHC & Immunofluorescence.
Immunofluorescence Analysis: A 1:200-1,000 dilution from a representative lot detected DLK1 in U251 cells.

Optimal working dilutions must be determined by end user.

Quality

Evaluated by Immunohistochemistry Analysis in human pancreas tissue.
