Specificity

Target description
Matrix remodeling-associated protein 8 (UniProt: Q9BRK3; also known as Dual Ig domain-containing cell adhesion molecule, DICAM, MXRA8, Limitrin) is encoded by the MXRA8 gene (Gene ID: 54587) in human. MXRA8/DICAM is a single-pass type I, homodimeric, membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 20-341), a transmembrane domain (aa 342-362), and a cytoplasmic domain (aa 363-442). It is highly expressed at the areas of cell-cell contacts. It is also detected in the soluble fraction of the nucleus. Its high expression is observed in the brain where it localizes to glia limitans formed by the end feet of astrocytes surrounding capillaries, and beneath the pia mater. It is also expressed in intercalated disks in the heart and has been detected in pancreatic -cells. It modulates the activity of various signaling pathways via binding to integrin V:β3 via its RGD motif (aa 252-254). It plays a role in cartilage formation where it promotes proliferation and maturation of growth plate chondrocytes and stimulates the formation of primary cilia in chondrocytes. MXRA8 also plays a role in the maturation and maintenance of the blood-brain barrier. It is reported to function as a cell trafficking molecule that is preferentially expressed by T helper 17 (TH17)-polarized CD4+ T lymphocytes. Its expression on circulating CD4+ T cells is increased in patients with active relapsing-remitting multiple sclerosis (RRMS) and in progressive forms of multiple sclerosis. Expression of its ligands also increases on the blood-brain barrier endothelium upon inflammation and in multiple sclerosis lesions. Storage and Stability
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
