Target description
Complement C4-B (UniProt: P01029; also known as C4B) is encoded by the C4b (also known as C4) gene (Gene ID: 12268) in murine species. The complement system provides innate defense against microbial infection and is a complement to antibody mediated immunity. It consists of about thirty-five interacting plasma and membrane associated proteins that contribute to host-defense. It contains several soluble factors that prevent spontaneous complement activation from occurring in solution, as well as several regulatory membrane-associated proteins that protect host cells from accidental complement attack. Complement C4 is a key component of the complement system and is one of chief constituents of innate immunity for immediate recognition and elimination of invading microbes. It plays an essential role for the functions of both classical and lectin complement pathways. It is a disulfide-bonded three-chain glycoprotein, consisting of an -chain (95 kDa), a β-chain (75 kDa), and a -chain (30 kDa). During complement activation, the -chain is cleaved by C1 into C4a and C4b, and C4b stays linked to the β- and -chains. Further degradation of C4b by C1 into the inactive fragments C4c and C4d blocks the generation of C3 convertase. Dysregulation of classic and lectin complement pathways where complement C4 plays a vital role can lead to complement-mediated autoimmune diseases. (Ref.: Wang, H., and Liu, M. (2021). Front.
