MHC class I polypeptide-related sequence A/MHC class I polypeptide-related sequence B (UniProt: Q29983/Q29980; also known as MIC-A/MIC-B) is encoded by the MICA/MICB (also known as PERB11.1/PERB11.2) gene (Gene ID: 100507436/4277) in human. MIC-A and B are closely related single-pass type I membrane proteins that act as stress-induced self-antigen that are recognized by gamma-delta T cells and serve as ligands for the KLRK1/NKG2D receptor. They are recognized by cytotoxic gamma-delta T cells expressing the TCR variable region V delta1. Under normal conditions expression of MIC-A and B is restricted mainly to the intestinal epithelium. However, their expression has been reported in tumor cells. They are proteolytically cleaved and released from the cell surface of tumor cells which impairs KLRK1/NKG2D expression and T-cell activation. MIC-A and B can be induced by heat shock and by exposure to DNA damaging conditions. They are synthesized with a signal peptide (aa 1-23 and 1-22, respectively for MIC-A and MIC-B), which are cleaved off in the mature form. Their mature forms contain an extracellular domain (aa 24-304 and 23-309), a transmembrane domain (aa 308-328 and 310-330), and a cytoplasmic domain (aa 329-383 and 331-383), respectively. Preparation Note
Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Evaluated by Flow Cytometry in HeLa cells.
