Histone acetyltransferase KAT2A (UniProt: Q92830; also known as EC: 2.3.1.48, General control of amino acid synthesis protein 5-like 2, Histone acetyltransferase GCN5, HsGCN5, Lysine acetyltransferase 2A, STAF97) is encoded by the KAT2A (also known as GCN5, GCN5L2, HGCN5) gene (Gene ID: 2648) in human. Histone acetyltransferase KAT2A belongs to the acetyltransferase family and is expressed in all tissues tested. However, its expression is highest in ovaries. It has three acetyl-CoA-binding regions (aa 579-581; 586-592; 617-620) and one bromodomain. It functions as a histone acetyltransferase (HAT) to promote transcriptional activation. Acetylation of histones gives a specific tag for epigenetic transcription activation. It displays significant histone acetyltransferase activity with core histones, but not with nucleosome core particles. KAT2A is also shown to acetylate non-histone proteins, such as CCAAT/enhancer-binding protein beta (CEBPB). In case of HIV-1 infection, histone acetyltransferase KAT2A is recruited by the viral protein Tat and it can regulates Tat′s transactivating activity and may help in inducing chromatin remodeling of proviral genes. Two isoforms of histone acetyltransferase KAT2A have been reported that are produced by alternative splicing. Isoform 2 lacks the first 410 amino acids.


Specificity

Clone 2GC-2C11 detects Histone acetyltransferase KAT2A in human and murine cells. It targets an epitope within 15 amino acids from the N-terminal region.

Immunogen

Ovalbumin-conjugated linear peptide corresponding to 15 amino acids from the N-terminal region of human Histone acetyltransferase KAT2A.

Application

Anti-KAT2A (GCN5), clone 2GC-2C11, Cat. No. MABE1791, is a highly specific mouse monoclonal antibody that targets Histone acetyltransferase KAT2A and has been tested in Western Blotting and Immunoprecipitation.
Western Blotting Analysis: 2 µg/mL from a representative lot detected KAT2A (GCN5) in 10 µg of NIH/3T3 cell lysate.

Immunoprecipitation Analysis: A representative lot detected KAT2A (GCN5) in Immunoprecipitation applications (Brand, M., et. al. (2001). EMBO J. 20(12):3187-96).

Western Blotting Analysis: A representative lot detected KAT2A (GCN5) in Western Blotting applications (Riss, A., et. al. (2015). J Biol Chem. 290(48):28997-9009; Helmlinger, D., et. al. (2006). PLoS Biol. 4(3):e67; Brand, M., et. al. (2001). EMBO J. 20(12):3187-96).

Quality

Evaluated by Western Blotting in HeLa nuclear extracts.

Western Blotting Analysis: 2 µg/mL of this antibody detected KAT2A (GCN5) in 10 µg of HeLa cells nuclear extract.
