General description

The protein known as Trop-2, or also known as cell surface glycoprotein Trop-2, Membrane component chromosome 1 surface marker 1 or Pancreatic carcinoma marker protein GA733-1 and encoded by the gene TACSTD2/GA733-1/M1S1/TROP2, is a protein that may function as a growth factor receptor and is expressed primarily in the placenta, lung and epithelial tissues but also found in certain cancers. The protein was first identified as a carcinoma antigen by the monoclonal GA733. It is a plasma membrane bound protein that transduces an intracellular calcium signal after binding unknown ligands. Mutations in Trop2 are associated with a rare eye disease leading to blindness. Trop-2 has also recently been shown to be important in regulating fibroblast motility and lung cell proliferation via the regulation of the ERK kinase pathway as well as several critical components of the cell migration apparatus.

Application

Immunocytochemistry Analysis: A representative lot detected Trop2 in MTE 4-14/T2 and MCF-7 cells (Prof. Saverio Alberti, Oncoxx Biotec.).

Flow Cytometry Analysis: A representative lot detected Trop2 in transfected cells, and in Skov-3 and HT-29 cancer cells (Prof. Saverio Alberti, Oncoxx Biotec.).

Inhibition Analysis: A representative lot inhibited tumor growth in DU145 prostate cancer cells, and in HT-29, HCT116 and Colo205 cells. (Note: When used together, Cat. No. MABC518, Anti-Trop2, clone 2EF, Azide Free and Cat. No. MABC519, Anti-Trop2, clone 2G10, Azide Free maximizes inhibition of tumor growth in vivo.) (Prof. Saverio Alberti, Oncoxx Biotec.).
This Anti-Trop2 Antibody, clone 2EF, Azide Free is validated for use in ICC, flow cytometry & inhibition for the detection of Trop2.

Quality

Evaluated by Immunocytochemistry in MET cells transfected with a vector expressing Trop2 or with an empty vector.

Immunocytochemistry Analysis: A 1:250 dilution of this antibody detected Trop2 in MET cells transfected with a vector expressing Trop2 and not in MET cells transfected with empty vector.

Trop-2 has limited presence on normal tissues but is highly expressed in diverse epithelial cancers. (E1)-3s is a T-cell-redirecting trivalent bispecific antibody (bsAb), comprising an anti-CD3 scFv covalently linked to a stabilized dimer of a Trop-2-targeting Fab using Dock-and-Lock. We
