General description

Apoptosis regulator Bax (UniProt: Q07812; also known as Bcl-2-like protein 4, Bcl2-L-4) is encoded by the BAX (also known as BCL2L4) gene (Gene ID: 581) in human. Bax is a pro-apoptotic member of the Bcl-2 family of proteins. It displays extensive homology with Bcl-2 and is variably expressed in different cells, usually at very low concentrations in normal cells. Upon induction of apoptosis Bax undergoes a conformational change and translocates from cytosol to mitochondrial membrane. Bax forms homodimers as well as heterodimers with Bcl-2 through common binding motif and the ratio of Bax to Bcl-2 determines the survival or death of cells during apoptosis. BAX is reported to interact with, and increase the opening of, the mitochondrial voltage-dependent anion channel (VDAC), which leads to the loss in membrane potential and the release of cytochrome c. Multiple alternatively spliced transcript variants, which encode different isoforms, have been reported for this gene. Clone 6A7 reacts with multiple isoforms of Bax.

Specificity

Clone 6A7 recognizes BAX in human cells. It targets an epitope with in 13 amino acids from the N-terminal region.

KLH-cojugated linear peptide coprresponding to 13 amino acids from the N-terminal region of human BAX.

Immunoprecipitation Analysis: A 1:250 dilution of this antibody detected Bax in THP-1 cell lysates.

Target description

~15 kDa observed; 21.18 kDa calculated. Uncharacterized bands may be observed in some lysate(s). Multiple isoforms for this target (8), ranging in MWs between 4-24 kDa, are reported. Most of the predicted MWs reside between 12-19 kDa Based on the sequence it is expected to react with isoforms 1,2,4,5, and 8.
