Specificity
Antibody MAB2575 was raised against mouse integrin VLA-5 (alpha5beta1), a fibronectin receptor. The antibody was prepared by fusion of spleen cells from a Fisher rat immunized with mouse embryonal carcinoma P19 cells with myeloma P3X63Ag9.653 cells. Specificity of the antibody was verified by its ability to immunoprecipitate alpha5beta1 heterodimers from (125)-I-surface labeled cells, reciprocal depletion of VLA-5 antigen from cell lysate with antiserum against the cytoplasmic domain of a5 integrin subunit, and immunoprecipitation of VLA-5 integrin from cells known to express this integrin. Cell lines tested to express alpha5beta1, include: P19 cells, mouse melanoma B16F1 cells, mouse squamous carcoma SCC cells and thymocytes. Characterization by immunoprecipitation was typically analyzed by SDS-PAGE using 6% gels under non-reducing conditions.
Immunogen
Mouse embryonal carcinoma P19 cells.
Application
Detect Integrin α5β1 using this Anti-Integrin α5β1 Antibody, clone BMC5 validated for use in FC, IP, IH.
Inhibition of cell adhesion to fibronectin: 10 μL/mL
Note that where cell adhesion to fibronectin involving other fibronectin receptors, such as VLA-4 (alpha4beta1) integrin, is involved, inhibition of cell adhesion by BMC5 alone may not be complete.
Immunoprecipitation from lysate of 10E6 cell equivalent: 2 μL
Flow cytometry: 10μL/mL
Immunohistochemistry on acetone-fixed frozen sections only; formalin fixation is not recommended.
Optimal working dilutions must be determined by end user.

Integrins are important adhesion molecules that regulate tumor and endothelial cell survival, proliferation and migration. The integrin alpha5beta1 has been shown to play a critical role during angiogenesis. An inhibitor of this integrin, volociximab (M200), inhibits endothelial cell growth and

Fibronectin (FN)-binding integrins control a variety of cellular responses through Rho GTPases. The FN-binding integrins, αvβ3 and α5β1, are known to induce different effects on cell morphology and motility. Here, we report that FN-bound αvβ3 integrin, but not FN-bound α5β1

Upon binding to the extracellular matrix protein, fibronectin, αV-class and α5β1 integrins trigger the recruitment of large protein assemblies and strengthen cell adhesion. Both integrin classes have been functionally specified, however their specific roles in immediate phases of cell attachment
