General description

Integrin proteins couple the ECM outside of a cell to the cytoskeleton inside of the cell. Which ECM protein binds and what signals are conveyed depend upon the particular makeup of the integrin pair being expressed. Functional integrin receptors are made up of two protein chains linked by disulfide bonds, an alpha chain and a beta chain. Integrin beta 4, also known as CD104, or GP160 is one of eight different beta chains, however beta 4 integrin is part of only one principal integrin receptor, integrin alpha6beta4. This receptor is expressed in endothelium, epithelium, and perineurium tissues of several organs as well as fibroblasts and Schwann cells but not in blood or lymphatic cells. Integrin alpha6beta4 is the laminin receptor and is critical for cellular polarity and motility.

SCC9 whole cells expressing Integrin B4.

Application

Immunohistochemistry Analysis: A representative lot detected Integrin B4 in acetone-fixed frozen human epithelial, tongue, foreskin, and ovary tissues. (Note: Not for use on formalin-fixed tissues.)

Inhibition Assay Analysis: A representative lot inhibits SCC9 cell adhesion to human and EHS laminin and laminin GD-2 peptides, but not to type IV collagen or fibronectin. It does not inhibit SKOV-3 cell adhesion to laminin, fibronectin or type IV collagen.

Flow Cytometry Analysis: A representative lot detected Integrin B4 in SCC9, SKOV-3, HUVEC cells. It also detected Integri B4 in RKO and K562 cells transfected for CD104 expression. It does not stain Integrin B4 in monocytes, neutrophils, lymphocytes, T cells, B cells or platelets.

Immunoprecipitation Analysis: A representative lot immunoprecipitated Integrin B4 from surface-biotinylated SCC9 cells. Immunoprecitated Integrin B4 was then observed at 66 kDa, 76 kDa, 120 kDa, 135 kDa and 200kDa (reduced), or 66 kDa, 110 kDa, 125 kDa, 130 kDa and 190kDa (non-reduced) in PAGE.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
