General description

Trypsin-1 (UniProt: P07477; also known as EC:3.4.21.4, Beta-trypsin, Cationic trypsinogen, Serine protease 1, Trypsin I) is encoded by the PRSS1 (also known as TRP1, TRY1, TRYP1) gene (Gene ID: 5644) in human. Trypsin is pancreatic serine protease that is synthesized as an inactive precursor known as trypsinogen that is processed through the Golgi apparatus and packaged into zymogen granules for secretion by exocytosis. Trypsin-1 is synthesized with a signal peptide (aa 1-15) and a propeptide (trypsin activation peptide; aa 16-23) that are subsequently cleaved off to generate the mature form that can be further cleaved and processed into alpha-trypsin chain 1 (aa 24-122) and alpha-trypsin chain 2 (aa 123-247). Trypsin can be present in a single-chain and a two-chain form, produced by proteolytic cleavage after arginine 122. Trypsin can also undergo sulfation at tyrosine 154 that increases its selectivity towards basic versus apolar residues at the P2′ position of inhibitors that bind in a substrate-like fashion. Trypsin cleaves peptide chains mainly at the carboxyl side of the amino acid lysine or arginine, except when either is followed by proline.

Specificity

Clone TRYP11-236.7 is a mouse monoclonal antibody that detects human Trypsin-1.

Immunogen

Purified human pancreatic trypsin.

Application

Anti-Trypsin, human pancreas, clone TRYP11-236.7, Cat. No. MAB1482-I, is a mouse monoclonal antibody that detects Trypsin and is tested for use in and ELISA.
Immunohistochestry/IH(P): {Solakidi, et al. 2003} 1:100-1:500; formalin fixed, paraffin embedded human tissues; 5 m serial sections; blocking solution 0.5% serum in PBS; overnight incubations; enzymatic detection.

ELISA: (Dilution is assay based)

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
