General description

Capsid protein VP1 (UniProt: Q6JC40) is encoded by the Cap gene in Adeno-associated virus 9 (AAV9). Adeno-associated viruses (AAV) are small non-enveloped DNA viruses that can serve as promising viral vectors for gene delivery, but can also elicits cellular and humoral immune responses. AAVs are reported to be nonpathogenic in humans and animals and display low immunogenicity. Following their attachment to cell surface receptor they are internalized via endocytosis and their intracellular trafficking is a rate-limiting, slow and inefficient process. AAVs contain an icosahedral capsid, which is 22 to 26 nm in diameter. They are shown to have a single-stranded DNA genome of approximately 4.7 kilobases that is flanked by two T-shaped inverted terminal repeats (ITRs). The 7C7 an epitope-tag antibody recognizes a basic ten amino acid epitope (QPAKKRLNFGQTGD) with high specificity. The basic nature of the BASE-tag epitope sequence distinguishes it from other commonly used epitope tags, such as Myc, EQKLISEEDL, FLAG, DYKDDDDK, HA, YPYDVPDYA, V5 etc. Clone 7C7 recognizes capsid proteins VP1 and VP2, but not VP3. Based on sequence identity of the epitope sequence, this antibody is expected to recognize VP1 and VP2 of AAV serotypes 1,2,3,6,7,8, and 9. (Ref.: Nonnenmacher, M., and Weber, T (2012). Specificity

Clone 7C7 is a mouse monoclonal antibody that detects various serotypes of Adeno-associated virus (AVV). It targets an epitope with in 14 amino acids from the N-terminal region.
