General description

Digitonin is a non-ionic steroid saponin derived from Digitalis purpurea. Historically, digitonin has been used to solubilize receptors and permeabilize membranes for sample preparation and analysis. Digitonin interacts with cholesterol creating a digitonin-cholesterol complex, which leads to subsequent removal of cholesterol and cellular membrane permeabilization. Therefore, cellular compartments composed of low amounts of cholesterol are protected from digitonin effects, particularly the nuclear membrane.

Due to digitonin′s mechanism of action, many other epigenetic applications utilize digitonin for nuclear extraction. For example, assay for Transposase-Accessible Chromatin with high throughput sequencing (ATAC-Seq) is a valuable method to map open chromatin sites and assess the epigenetic landscape. In combination with other detergents, a low concentration of digitonin is employed to isolate the nuclear pellet followed by digestion of the nuclei with an increased concentration of digitonin5. In our ChIC (Chromatin Immuno-Cleavage) Kits (part #s: CHR100-102), digitonin is utilized for antibody targeting of micrococcal nuclease for chromatin tagging.
