Application

Collagen IV from the human placenta is suitable for use as a coating material for cell culture studies and the formation of collagen gels. Human Collagen Type IV has been used: to prepare collagen gel for the invasion assay to provide a supportive matrix for assessing the invasive capacity of monocytes in a cell culture systemas an extracellular matrix (ECM) protein in microarray analysis, to coat nitrocellulose film slidesto coat the microchannels of the P96-O2 plate, and provide a surface for the adhesion and growth of endothelial cells (hMVECs) and renal proximal tubular epithelial cells (hRPTECs) in a co-culture system

Biochem/physiol Actions

Human collagen type IV is associated with Alport and Goodpasture′s syndrome, as well as several rheumatological and dermatological diseases including, acquired epidermolysis bullosa.

General description

Human collagen type IV is a major structural element predominantly found in the basement membrane of the skin. It is a native triple helix and heterotrimeric protein containing two α1-like and one α2-like genes. Type 4 collagen forms a sheet-like extracellular matrix in multicellular tissues that exists around certain cell types like adipocytes, skeletal muscle cells, heart muscle cells, and smooth muscle cells. Human Collagen IV is extracted from washed dissected tissue into dilute acetic acid after mild pepsin treatment. The Collagen type 4 was purified by using differential salt precipitation. Purity and retention of the native helical structure were controlled by SDS-PAGE and by reaction with anti-collagen type-specific monoclonal antibodies.
