General description

Pancreatic triacylglycerol lipase (EC 3.1.1.3; UniProt P35638; also known as Pancreatic lipase, PL, PTL, Triacylglycerol acylhydrolase) is encoded by the PNLIP (Also known as PNLIPD) gene in human (Gene ID 5406). PTL is a pancreas synthesized carboxyl esterase that hydrolyzes insoluble, emulsified triglycerides (TAGs). The dietary TAGs are partially digested by the gastric lipase in the stomach. The emulsion particles then mix with digestive enzymes from the pancreas in the duodenum where digestion proceeds. The pancreas secretes multiple lipases, including pancreatic triglyceride lipase (PTL), carboxyl ester lipase (CEL), and pancreatic lipase-related protein-1 and -2 (PLRP1 & PLRP2). Unlike PLRP2, PLRP1 exhibits little activity against triglycerides and no measurable activity against phospholipids, galactolipids or cholesterol esters. Experimental evidences suggest that PLRP1 plays a negative regulatory role against PLT–colipase-mediated dietary triglyceride digestion. Pancreas-derived lipases are essential for the efficient digestion of dietary TAG. Pancreatic insufficiency or exocrine pancreas failure results in steatorrhea or steatorrhoea, where increased fat excretion is seen in fecal samples.

Reacts with pancreatic triglyceride lipase (PTL), pancreatic lipase-related protein-1 & -2 (PLRP1 & PLRP2), and the truncated PLRP2 W340X mutant.

Western Blotting Analysis: A representative lot detected recombinant mouse pancreatic triglyceride lipase (PTL/PNLIP), as well as endogenous pancreatic lipase-related protein-2 (PLRP2) in mouse pancreas (Xiao, X., et al. (2011) J. of Lipid Res. 52(5):982-990).
Western Blotting Analysis: A representative lot detected both wild-type and W340X mutated recombinant human pancreatic lipase-related protein-2 (PLRP2) in culture media and lysates from transfected 293T and COS-7 cells (Xiao, X., et al. (2011) J. Biol. Chem. 286(30):26353-26363).
Neutralizing Analysis: A representative lot inhibited colipase-dependent pancreatic lipase-related protein-2 (PLRP2) enzymatic activity in pancreas extracts from 4-day old mice (D′Agostino D, and Lowe ME. (2004) J. Nutr. 134(1):132-134).

Quality

Evaluated by Western Blotting in human pancreas tissue lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antibody detected Pancreatic Lipase/PNLIP in 10 µg of human pancreas tissue lysate.

Target description

~53 kDa observed. 51.1 kDa (Human PTL, calculated), 51.4 kDa (Mouse & Rat PTL, calculated), 51.8/51.9 kDa (Human PLRP1/2, calculated), 52.7/52.6 kDa (Mouse PLRP1/2, calculated), 52.4/52.5 kDa (Rat PLRP1/2, calculated). Target band may migrate as a doublet on gel due to glycosylation.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
