General description

GBP5, also known as GBP-TA antigen, or GTP-binding protein 5, or Guanine nucleotide-binding protein 5 or GBP-5, and encoded by the gene GBP5, is a protein that plays a role in the inflammatory response to pathogens and may also play a role in apoptosis. GBP5 stimulates cryopryrin (NLRP3 protein) assembly of the intracellular inflammasome, a multiprotein oligomer consisting of caspase 1, PYCARD, NALP and sometimes caspase 5. Inflammasomes act as sensory complexes that alert the immune system to the presence of infection or tissue damage. Nucleosomes and GBP5 are expressed in myeloid cells and are a component of the innate immune system; GBP5 acts as key rheostat for activation and assembly of the inflammasomes. Additionally, GBP5 is a GTPase that homodimerizes upon GTP-binding; dimerization is required for the second hydrolysis step from GDP to GMP. GBP5 is localized to the cytoplasm and intracellular membranes via its lipid-anchor sequences.

Western Blotting Analysis: A representative lot from an independent laboratory detected GBP5 in differentiating THP-1 cells which were untreated or treated with LPS alone, or LPS/IFN-gamma (Shenoy, A. R., et al. (2012). Science. 336(6080):481-485.).

Evaluated by Western Blotting in K562 cell lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected GBP5 in 10 µg of K562 cell lysate.

Target description

~67 kDa observed. A second isoform is known to exist at 55 kDa Uncharacterized band(s) may be observed in some cell lysates.
