General description

Heat shock protein 75 kDa, mitochondrial (UniProt: Q12931; also known as HSP 75, TNFR-associated protein 1, Tumor necrosis factor type 1 receptor-associated protein, TRAP-1) is encoded by the Trap1 (also known as Hsp75) gene (Gene ID: 287069) in human. TRAP-1 is a mitochondrial protein that serves as a chaperone with ATPase activity. Its ATPase activity is inhibited by small molecule pocket antagonists, such as geldanamycin or radicicol. TRAP-1 is expressed in skeletal muscle, liver, heart, brain, kidney, pancreas, lung, placenta and bladder. Higher expression levels are observed in several cancer cell lines. However, bladder and renal cancers display reduced levels of TRAP-1. TRAP-1 is involved in maintaining mitochondrial function and polarization, likely through stabilization of mitochondrial complex I. Two isoforms of TRAP-1 have been reported that are produced by alternative splicing. It is synthesized with a transit peptide (aa 1-50) that is subsequently cleaved off. TRAP-1 is reported to protect cells against mitochondrial apoptosis and helps cells to cope with oxidative stress. (Ref.: Altieri, DC et al. (2012). Biochim. Biophys. Acta 1823(3); 767-773).


Specificity

This rabbit polyclonal antibody detects Tumor necrosis factor type 1 receptor-associated protein (TRAP-1; HSP75) in human cells. It targets an epitope within 12 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linenar peptide corresponding to 12 amino acids from the C-terminal half of human TRAP-1 (HSP75). The immunogen sequence is conserved in both isoforms.

Application

Anti-TRAP-1 (HSP75), Cat. No. ABS1489, is a rabbit polyclonal antibody that detects TRAP-1 (HSP75) and has been tested for use in Immunohistochemistry (Paraffin) and Western Blotting.
Western Blotting Analysis: 2 µg/mL from a representative lot detected TRAP-1 (HSP75) in 10 µg of HeLa cell lysate.

Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected TRAP-1 (HSP75) in human brain, human tonsil, and human prostate cancer tissues.

Quality

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected TRAP-1 (HSP75) in 10 µg of HepG2 cell lysate.

Target description

~75 kDa observed; 80.11 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
