General description

The amino acid Hypusine is a unique amino acid and a critical component of only one cellular protein, the eukaryotic translation initiation factor 5A (eIF-5A, old terminology eIF-4D). Hypusine is formed post-translationally in two enzymatic steps that modify a specific lysine in the precursor protein of eIF-5A. Hypusine is produced soon after the translation of eIF-5A and the modification is essentially irreversible. Hypusine is found in all eukaryotes and archaebacteria but not in eubacteria, and the high conservation of the amino acids surrounding the key lysine point to its importance in eukaryotic biology. The expression of eIF-5A is essential as is the lysine for eIF-5A function. eIF-5A is needed for the initiation of translation and other important processes such as responses to cellular stress, and nucleocytoplasmic shuttling of specific mRNAs. Without an active Hypusine cell proliferation is inhibited, thus Hypusine and its biosynthesis are targets of anti-cancer therapies.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage. Do not store in -80°C.
