General description

BAG family molecular chaperone regulator 3 (UniProt Q9JLV1; also known as BAG-3, Bcl-2-associated athanogene 3, Bcl-2-binding protein Bis) is encoded by the Bag3 (also known as Bis, MNCb-2243) gene (Gene ID 29810) in murine species. Bcl-2-associated athanogene 3 (BAG3) is a stress-activated protein abundantly expressed in the heart, skeletal muscles, and many cancers. BAG3 is involved in protein quality control (PQC) by serving as a co-chaperone for both constitutive and inducible heat shock proteins (Hsc/Hsp), as well as in mediating cellular anti-apoptotic activity via Bcl2 binding. BAG3 also plays a role in sarcomere stability by regulating filamin clearance and production, as well as by interacting with actin capping protein beta 1 (CapZβ1) that binds the barbed end of actin and prevent actinn fibers from disssociating into actin monomers. While BAG3 is found in the cytoplasm of neonatal myocytes and involved in PQC and apoptosis, BAG3 is localized predominantly in the sarcolemma and t-tubules of adult cardiac myocytes, where it modulates myocyte contraction in response to β-adrenergic receptor signaling. Bag3-knock mice die 4 weeks after birth due to non-inflammatory myofibrillar degeneration. Human BAG3 gene mutations are found in childhood-onset muscular dystrophy, in families with dilated cardiomyopathy, as well as in cases of idiopathic dilated cardiomyopathy.

Specificity

This chicken polyclonal antibody detects mouse BAG3 by targeting a C-terminal region sequence. Rat species cross-reactivity is likely, but has not been confirmed.

Immunogen

KLH-conjugated linear peptide corresponding a sequence near the C-terminus of mouse BAG3.

Quality

Evaluated by Western Blotting in mouse heart tissue lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected BAG3 in 10 µg of mouse heart tissue lysate.

Target description

~80 kDa observed. 61.73 kDa calculated (Met1 removed). the larger-than-calculated band size is consistent with that reported in the literature (Rasati, A., et al. (2015). Nat. Commun. 6:8695). Uncharacterized bands may be observed in some lysate(s).
