General description

Brain-derived neurotrophic factor (UniProt P23560; also known as Abrineurin, BDNF, Neurotrophin) is encoded by the BDNF (also known as BULN2) gene (Gene ID 627) in human. BDNF is a member of the NGF family of neurotrophic factors (also named neurotrophins) that are required for the differentiation and survival of specific neuronal subpopulations in both the central as well as the peripheral nervous system. The neurotrophin family is comprised of at least four proteins including NGF, BDNF, NT3, and NT4/5. BDNF mediates its neurotrophic properties by signaling through the high affinity cell surface receptor gp145/trkB. BDNF is initially produced with a signal peptide (a.a. 1-18) and a propeptide (a.a. 19-128) sequence, the removal of which yields the mature BDNF (a.a. 129-247).

Specificity

This polyclonal antibody detects prepro- and pro-BDNF, as well as prepro-/pro-BDNF-derived N-terminal fragments that contain the propeptide sequence (a.a. 19-128), but not mature BDNF or C-terminal fragments without the propeptide sequence. No cross-reactivity to other proneurotrophins.

Target description

~35 kDa (glycosylated prepro-/pro-BDNF), ~50-65 kDa (glycosylated prepro-/pro-BDNF dimer), and ~26-28 kDa (non-glycosylated prepro/pro-BDNF or glycosylated prepro-/pro-BDNF N-terminal fragments) observed. Target bands appear larger than the calculated molecular weights of 27.82/28.87/29.82/37.12/31.12 kDa (human prepro-BDNF isoform 1/2/3/4/5) and 25.77/26.82/27.77/35.07/29.06 kDa (human pro-BDNF isoform 1/2/3/4/5) due to glycosylation and/or dimerization. Uncharacterized band(s) may appear in some lysates.

Physical form

Lyophilized from 10 mM PBS. Reconstitute with 20 μL of sterile distilled water.

Storage and Stability

Maintain lyophilized material at -20°C for up to 6 months after date of receipt. After reconstitution maintain at -20°C undiluted for up to 6 months. Avoid repeated freeze/thaw cycles. Glycerol (ACS grade or better) can be added (1:1) for additional stability.
