General description

Beta-lactamase TEM (EC 3.5.2.6; UniProt P62593; also known as IRT-4, Penicillinase, TEM-1, TEM-16/CAZ-7, TEM-2, TEM-24/CAZ-6, TEM-3, TEM-4, TEM-5, TEM-6, TEM-8/CAZ-2) is encoded by the bla (also known as blaT-3, blaT-4, blaT-5, blaT-6) gene (Gene ID 3244915) in E. coli. TEM-1 belongs to the class-A beta-lactamase family. Beta-lactamases produced by bacteria provide resistance to beta-lactam antibiotics, such as penicillins and cephamycins by breaking open the beta-lactam ring. Over 200 beta-lactamases have been reported that are classified into four main groups (A, B, C, and D) and eight subgroups according to their functional and structural characteristics. The TEM-1 is predominant plasmid-mediated beta-lactamase of gram-negative rods. Six different nucleotide sequences, called blaTEM-1a, blaTEM-1b, blaTEM-1c, blaTEM-1d, blaTEM-1e, and blaTEM-1f, have been described to codify the same TEM-1 beta-lactamase. Ref.: Briñas, L., et al. (2002). Antimicrob. Agents Chemother. 46, 103156-103163.

Specificity

This polyclonal antibody detects E. coli beta-lactamase.

Immunogen

GST-tagged full-length E. coli beta-lactamase, including the signal peptide sequence.

Application

Detect beta-lactamase using this rabbit Anti-Beta Lactamase polyclonal antibody, Cat. No. AB3738-I, validated for use in Western Blotting.

Quality

Evaluated by Western Blotting in lasate from beta-lactamase-transformed E.coli.

Western Blotting Analysis: A 1:100,000 dilution of this antibody detected GST-tagged full-length E. coli beta-lactamase in 0.5 µg of lysate from transformed E. coli.

Target description

31.52/28.91 kDa (with/without signal peptide a.a. 1-23) calculated.

Linkage

Replaces: AB3738

Cotranslational insertion of selenocysteine (Sec) proceeds by recoding UGA to a sense codon. This recoding is governed by the Sec insertion sequence (SECIS) element, an RNA structure on the mRNA, but size, location, structure determinants, and mechanism differ for Bacteria
