General description
Neuroendocrine convertase 2 (EC 3.4.21.94; UniProt P16519; also known as KEX2-like endoprotease 2, NEC 2, PC2, Prohormone convertase 2, Proprotein convertase 2) is encoded by the PCSK2 (also known as NEC2) gene (Gene ID 5126) in human. Neuroendocrine convertase 2/PC2 belongs to the family of subtilisin-related proteolytic enzymes involved in the proteolytic cleavages of peptide hormone precursors. For example, during the multistep processing of the peptide precursor proopiomelanocortin (POMC), PC2 catalyzes the cleavage of adrenocorticotropic hormone 1–39 (ACTH) into adrenocorticotropic hormone 1–14, which is then further processed to produce α-melanocyte-stimulating hormone (α-MSH). PC2 is produced initially in a prepro-form with a signal peptide (a.a. 1-25) and a propeptide (a.a. 26-109) sequence, the removal of which yields the mature 529-amino acid enzyme (a.a. 110-638). The neuroendocrine protein 7B2 blocks proPC2 from forming unactivatable aggregates and is absolutely required for processing proCP2 into active enzyme. Newly synthesized proPC2 is N-glycosylated in the ER and undergoes rate-limiting folding process before it can bind 7B2 and be transported to the TGN/ secretory granules for maturation by sulfation and proteolytic cleavage.
Specificity
This polyclonal antibody detects the ~68 kDa secreted mature PC2 in mouse cerebellum lysate by Western blotting. Expected to recognize also the Prepro- and pre-forms of all three spliced human isoforms reported by UniProt (P16519).

Target description
~68 kDa calculated. Target band appears larger than the calculated molecular weight of the mature form (58.28 kDa/mouse & 58.00 kDa/human) due to posttranslational glycosylation and sulfation. 67.97/63.72/68.12 kDa (human isoform 1/2/3 proforms), 70.57/66.31/68.58 kDa (human isoform 1/2/3 prepro-forms) calculated.
