General description

Tryptophan hydroxylase (TPH) is an enzyme involved in the synthesis of the neurotransmitter serotonin. It is responsible for addition of the -OH group (hydroxylation) to the 5 position to form the amino acid 5-hydroxytryptophan (5-HTP), which is the initial and rate-limiting step in the synthesis of the hormone serotonin. It is also the first enzyme in the synthesis of melatonin.
Immunogen
Recombinant rabbit tryptophan hydroxylase, isolated as inclusion bodies from E. coli and purified by preparative SDS-PAGE.

Detect Tryptophan Hydroxylase using this Anti-Tryptophan Hydroxylase Antibody validated for use in IH & WB.
Immunohistochemistry(peroxidase):
Performed on formaldehyde fixed tissue of a previous lot. Useful for cell body but not fiber staining.

Biochem/physiol Actions
Reacts with human and rat brain tissues by immunoblotting and immunohistochemistry. Based on the relatively high degree of homology of tryptophan hydroxylase, the antibody should cross-react with other mammalian species.

Based on the relatively high degree of homology of tryptophan hydroxylase, the antibody should cross-react with other mammalian species. Recently studies have indicated that AB1541 can cross react with Tyrosine hydroxylase on western blots, weakly. Cross reactivity in the native state is untested.
Physical form
Purified sheep polyclonal liquid in 150 mM NaCl, 10 mM HEPES (pH 7.5), 100 μg/mL BSA and 50% glycerol.
Preparation Note
Stable for 6 months at -20ºC to -80ºC in undiluted aliquots from date of receipt.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol-containing solutions to become frozen during storage.
