General description

An internally quenched, highly cathepsin S-selective, fluorogenic peptide substrate that is efficiently cleaved by CatS (kcat/Km = 1.018 µM-1 S-1), but not by Cathepsin B & L (kcat/Km = 0.004 and 0.032 µM-1 S-1, respectively). Studies using CatS-specific inhibitor LHVS reveal that the substrate is not cleaved by other proteolytic enzymes, including CatB, CatH, CatL, and CatX, present in endosomal extracts from human B cell line WT100, dendritic cells, and macrophages (excitation maximum: 340 nm; emission maximum: 405 nm).
