TANK-binding kinase-1 (TBK1), also known as NF-κB activating kinase (NAK), is a serine-threonine kinase from the IκB kinase (IKK) family. It is encoded by tbk1 gene and is ubiquitously expressed in various tissues.

Biochem/physiol Actions

TANK-binding kinase-1 (TBK1) is mainly involved in activating interferon regulatory transcription factor 3/7 (IRF 3/7) and nuclear factor (NF-κB) signaling pathways, which are essential for regulating the production of type I interferons (IFN), including IFN-α and IFN-β and the activation of innate immunity. In addition, TBK1 also mediates various other cellular processes including autophagy, proliferation, immune response/inflammation survival, insulin signaling, and metabolism. TBK1 plays a significant role in cancer pathogenesis. In cancer cells, it enhances tumor growth by activating proliferation and survival signaling, promoting tumor-promoting cytokine production, and facilitating the secretion of immunosuppressive inflammatory cytokines. Overexpression of tbk1 gene is observed in various cancers including non-small cell lung cancer (NSCLC), pancreatic ductal adenocarcinoma (PDA), cholangiocarcinoma, clear cell renal cell carcinoma (ccRCC), adult T-cell leukemia, melanoma, esophageal cancer, and breast cancer.
