General description
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.
Specificity
Broad species cross-reactivity is expected based on sequence homology.
Recognizes monomethylated histone H3 (Lys9), Mr ~17 kDa.
Immunogen
Epitope: Lys9
KLH-conjugated, synthetic 2X-branched peptide containing the sequence …ARmeKST… in which meK corresponds to monomethyl-lysine at residue 9 of human histone H3.
Application
Anti-monomethyl-Histone H3 (Lys9) Antibody is a Rabbit Polyclonal Antibody for detection of Histone H3 monomethylated on lysine 9 (H3K9me1). This purified antibody is specificity verified by dot blot (DB), published in peer reviewed journals, and has been validated in ICC, PIA & WB.
Peptide Competition:
Specificity confirmed by the ability of 10 μM of the immunizing peptide to abolish detection of histone H3 in immunoblot analysis of HeLa acid extracts (Figure B, Lane 2). No signal reductions were detected with preincubation with 10 μM di- or trimethyl-lysine 9 or di- or trimethyl-lysine 27 modified peptides. Some signal reduction detected after preincubation with 10 μM monomethyl-lysine 27 modified peptide (Figure B, Lane 5, page 2).
Immunocytochemistry & Dot Blot: Reported by an independent laboratory1-4.

Quality
Routinely evaluated by Western Blot on acid extracted proteins from HeLa cells, but did not detect unmethylated recombinant Histone H3.
Western Blot Analysis:
0.1-0.2 μg/mL had detected monomethylated histone H3 in acid extracted proteins from HeLa cells, but did not detect unmethylated recombinant Histone H3 (Catalog # 14-411).

Post-translational modifications of histones by chromatin modifying enzymes regulate chromatin structure and gene expression. As deregulation of histone modifications contributes to cancer progression, inhibition of chromatin modifying enzymes such as histone demethylases is an attractive therapeutic strategy to impair cancer

Methylation of DNA and lysine 9 of histone H3 (H3K9) are well-conserved epigenetic marks for transcriptional silencing. Although H3K9 methylation directs DNA methylation in filamentous fungi and plants, this pathway has not been corroborated in mammals. G9a and GLP/Eu-HMTase1 are
