Gene Information

General description

Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H4 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. Acetylation of histone H4 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).

Specificity

This antibody is expected to demonstrate broad species cross-reactivity.
This antibody recognizes the N-terminus of Histone H4 acetylated at Lys5.

Immunogen

Epitope: N-terminus
KLH-conjugated linear peptide corresponding to the N-terminus of Histone H4 acetylated at Lys5.

Application

Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected acetyl-Histone H4 in HeLa and A431 cells.

Dot Blot Specificity Analysis: A representative lot detected acetyl-histone H4 (Lys5) peptides, but not unacetylated-histone H4 (Lys4) peptides or other peptides corresponding to modified histones.

Chromatin Immunoprecipitation Analysis: 5 µL from a representative lot immunoprecipitated acetyl-Histone H4 in HeLa S3 chromatin.

Use Anti-acetyl-Histone H4 (Lys5) Antibody (Rabbit Polyclonal Antibody) validated in WB, DB, ICC, ChIP-seq, ChIP to detect acetyl-Histone H4 (Lys5) also known as H4K5Ac, Histone H4 (acetyl K5).

Quality

Evaluated by Western Blot in HeLa acid extract.

Western Blot Analysis: A 1:2,000 dilution of this antibody detected acetyl-Histone H4 in 10 µg of HeLa acid extract.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

Control
HeLa acid extract
