General description

In humans, phosphoserine (PS) is present in the intracellular membranes and grey matter. It is localized to the nuclear envelope, endoplasmic reticulum, Golgi apparatus, outer mitochondrial membranes, plasma membranes and myelin.

Application

17:0-20:4 PS or 1-heptadecanoyl-2-(5Z,8Z,11Z,14Z-eicosatetraenoyl)-sn-glycero-3-phospho-L-serine has been used as an internal standard:
  • in lipid extraction from isolated pure mitochondria for mass spectrometry (MS) analysis
  • for lipid quantification/analysis using liquid chromatography (LC)-MS
  • for phospholipid quantification by MS
  • in lysophospholipid acyltransferases (LPLAT) assay
  • in the preparation of total lipid extracts from lysosome samples by tert-butyl methyl ether (MTBE) lipid extraction protocol for analysis by LC-MS
  • for ratiometric quantification of phosphatidylserine (PS) using LC-MS

Biochem/physiol Actions

Phosphoserine (PS) maintains the nerve cell membrane and myelin sheath. It is found to be associated with cognitive disability in Alzheimer′s disease.

Neurons from the adult central nervous system (CNS) demonstrate limited mRNA transport and localized protein synthesis versus developing neurons, correlating with lower regenerative capacity. We found that deimination (posttranslational conversion of protein-bound arginine into citrulline) undergoes upregulation during early neuronal

Cellular membranes contain glycerophospholipids, which have important structural and functional roles in cells. Glycerophospholipids are first formed in the de novo pathway (Kennedy pathway) and are matured in the remodeling pathway (Lands' cycle). Recently, lysophospholipid acyltransferases functioning in Lands' cycle
