General description

7β-hydroxycholesterol (7βOHC) is an oxidized derivative of cholesterol. 7β-hydroxycholesterol-d7 is the deuterated form of 7β-hydroxycholesterol.

Application

7β-hydroxycholesterol-d7 has been used:
  • as cholesterol oxidation product for mass spectroscopy analysis
  • as an internal standard for spiking white adipose tissue and plasma samples for mass spectroscopy analysis
  • as an internal standard working solution in gas chromatography- triple-quadrupole mass spectrometry
  • as an isotopically labeled internal standard in tandem mass spectroscopy analysis for plasma sterol quantification

Biochem/physiol Actions

7β-hydroxycholesterol-d7 (7β-OHC) along with 7-ketocholesterol (7-KC) plays a key role in atherogenesis. It is distributed extensively in the arterial compartment of apo- E knockout mice. 7βOHC induces apoptosis via activation of glycogen synthase kinase-3 (GSK3). It also triggers reactive oxygen species (ROS) resulting in mitochondrial dysfunction. 7βOHC favors demyelination and is a potential oxidized oxysterol in X-linked adrenoleukodystrophy (X-ALD) disorder.

Obesity may not be consistently associated with metabolic disorders and mortality later in life, prompting exploration of the challenging concept of healthy obesity. Here, the consumption of a high-fat/high-sucrose (HF/HS) diet produces hyperglycaemia and hypercholesterolaemia, increases oxidative stress, increases endotoxaemia

Oxysterols are important intermediates in numerous metabolic and catabolic pathways and their biological significance is also proven. The present paper describes a reliable and short liquid chromatography-high-resolution mass spectrometry method (LC-MS/HR-MS) for the quantification of 8 different oxysterols (24(S)-hydroxycholesterol, 25-hydroxycholesterol
