一般描述

TMB底物是辣根过氧化物酶-HRP的显色底物。

应用

TMB底物可用于利用辣根过氧化物酶结合物进行的ELISA程序。TMB底物会形成可溶的蓝色反应产物,可通过370或655 nm处的吸光度测定。该试剂可与样品中的亚硝酸盐反应,形成紫色的偶氮产物,可在450 nm处测量其吸光度。该反应可利用酸终止,形成黄色反应产物,并可通过450 nm处的光强精确测定。

我们的TMB底物使用方法:
  • 我们的TMB底物以即用型过氧化物酶底物的形式提供,其中的TMB溶于弱酸性缓冲液中。与过氧化物酶反应之前,底物溶液应呈无色至浅蓝绿色。与过氧化物酶反应时,底物系统形成蓝色反应产物。
  • 加入酸终止反应后,测量TMB底物在450 nm处的光强。
  • 我们的TMB底物推荐用于ELISA(微孔)程序,且已应用在各种测定方案中,如作为辣根过氧化物酶抗体偶联物的底物,用于检测亨德拉病毒糖蛋白、十四糖、sIgA和嗜中性白细胞胞外阱(NETs)。
  • 不建议采用3,3′,5,5′-四甲基联苯胺(TMB)液体底物系统用于膜应用。由于TMB底物会产生可溶性反应产物,因此不建议用于组织化学或印迹分析。
注意:TMB底物为光敏材料,应避免接触直射阳光或紫外线。

      No hydrogen peroxide should be added to product T0440. The product, which contains  a peroxidase substrate,  already contains hydrogen peroxide.  This solution is ready to use for ELISA assays.

    4. What is the concentration of the TMB and what is the solvent in Product T0440, 3, 3´,5,5´-Tetramethylbenzidine (TMB) Liquid Substrate System for ELISA?

      The concentration of TMB in this formulation and the identity of the solvent are considered proprietary information. The product is a ready-to-use peroxidase substrate containing TMB in a mildly acidic buffer.

    5. Do I need to dilute Product T0440, 3, 3´,5,5´-Tetramethylbenzidine (TMB) Liquid Substrate System for ELISA?

      No, the product should not be diluted.. The product is ready-to-use for ELISA applications. If the intensity of the reaction needs to be reduced, it is recommended that the antibodies or  conjugates be diluted. Usage information  is described on our data sheet.

    6. What is the difference between products T0440, 3, 3´,5,5´-Tetramethylbenzidine (TMB) Liquid Substrate System for ELISA,  and T8665, 3,3´,5,5´-Tetramethylbenzidine (TMB) Liquid Substrate System?

      Both products are comparable and either one can be used in ELISA procedures. The formulations which are proprietary may have some slight differences.
