一般描述

玻连蛋白是肝脏中合成的分泌型糖蛋白。它也由呼吸道上皮细胞分泌，存在于健康的肺部。主要以单体形式循环，但也可改变构象形成二硫键连接的多聚体。玻连蛋白基因由约4.5–5 kb碱基对组成，含有8个外显子和7个内含子。
重组人玻连蛋白含有459个氨基酸。存在两种形式：单链和截短形式，后者由内源性切割形成二硫键连接的双链。该重组玻连蛋白化学成分明确，可用作人胚胎干细胞更新培养基的基质成分。

应用

人玻璃粘连蛋白已被用于研究细胞外基质 (ECM) 配体对 C6 胶质瘤细胞的作用。它还被用作促进附着的蛋白来覆盖细胞板。

生化/生理作用

多聚玻连蛋白可高效结合并加入细胞外基质。在基质内，玻连蛋白通过结合各种整合蛋白及其他蛋白聚糖支持细胞黏附。与整合蛋白αvβ3的相互作用可增强缺氧耐受性和肿瘤细胞侵袭。还与各种癌症进展有关。还参与创伤修复，也会调控补体系统。此外，玻连蛋白还是肝脏、心脏和肾脏中的促纤维化活性标志物。

重悬

先离心，再开瓶。在水中重悬至浓度为0.1-1.0 mg/ml。请勿涡旋。该溶液可在2-8C°下储存长达1周。对于长期储存，建议在含有载体蛋白（如0.1%BSA）的缓冲液中进一步稀释，分装为工作体积并在-20°C至-80°C下储存。
