胃蛋白酶是一种用于消化蛋白的肽酶，通常用于从抗体制备Fab片段。来自猪胃粘膜的胃蛋白酶已用于水解小鼠中的干燥宫颈样品。产品P7125以粉末形式提供。产品P7125已被用于使肾细胞中的DNA变性[3] 并且在进行EGFR染色之前，对来自肛管癌（ACC）的活检病理学样品进行消化[4]。

生化/生理作用

优先裂解：P1 和 P1′ 位的疏水性和芳族残基。降解胰岛素 β 链中的 Phe-Val、Gln-His、Glu-Ala、Ala-Leu、Leu-Tyr、Tyr-Leu、Gly-Phe、Phe-Phe 和 Phe-Tyr 键

它不会在缬氨酸、丙氨酸或甘氨酸连接键上进行裂解。[5]ZL-酪氨酰-L-苯丙氨酸、ZL-谷氨酰-L-酪氨酸或ZL-甲硫氨酰-L-酪氨酸可用作胃蛋白酶消化的底物。[5][6]胃蛋白酶可被几种含苯丙氨酸的肽抑制。[7]

胃蛋白酶可水解肽键，但不可水解酰胺或酯键。胃蛋白酶可在具有芳香酸的肽的肽键任一侧进行切割。当含硫氨基酸接近肽键时，它们会增加肽的水解敏感性。胃蛋白酶优先在苯丙氨酸和亮氨酸的羧基侧以及谷氨酸残基的羧基侧进行切割。其可切割胰岛素β链中的Phe-Val、Gln-His、Glu-Ala、Ala-Leu、Leu-Tyr、Tyr-Leu、Gly-Phe、Phe-Phe和Phe-Tyr键
胃蛋白酶是胃中产生的主要蛋白水解酶。它可通过内部肽键的切割消化蛋白质。[8]

单位定义

在pH2.0，37℃条件下，一个单位每分钟可产生0.001ΔA₂₈₀（用血红蛋白作为底物，对TCA可溶性产物进行测定）。（终体积= 16 mL。光路 = 1cm。)

分析说明

最适pH值为2-4。在4 M尿素和3 M盐酸胍中可保持活性。在60°C时稳定。胃蛋白酶在pH 8.0-8.5下发生不可逆灭活。

蛋白浓度由E1％/ 280进行确定
