Biochem/physiol Actions

作用机制：制霉菌素通过与真菌细胞膜中的麦角甾醇结合，增加膜的渗透性和产生基于制霉菌素/麦角甾醇的离子通道起作用。它还可作为脂筏抑制剂和膜相关胆固醇。

抗菌谱：制霉菌素可对抗真菌、酵母和霉菌。

对严重烧伤患者的研究表明，制霉菌素粉治疗对大规模的血管侵入性真菌感染有效，对伤口愈合没有副作用。

Disclaimer

制霉菌素的组织培养基溶液在37°C下可稳定保存三天。在中等碱性介质中也很稳定。溶液和水悬液在制备后不久就开始失去活性，热、光和氧气会加速分解。如果在pH 7下加热至100°C，水悬液可稳定保存10 min。

General description

制霉菌素是多烯大环内酯类抗真菌药，它是由诺尔斯氏链霉菌 ATCC 11455 生产的聚酮化合物。该抗生素含有一个大内酯环，其天然合成是通过乙酰辅酶 A 起始的，然后缩合三个乙基丙二酰辅酶 A 和 15 个丙二酰辅酶 A 延伸部分。

保存于密封容器内，置于干燥通风处。在惰性气体下储存。对空气和光敏感。置于干燥处保存。

Preparation Note

在28°C，制霉菌素可溶于甲醇（11.2 mg/mL）、乙醇（1.2 mg/mL）、四氯化碳（1.23 mg/mL）、氯仿（0.48 mg/mL）、苯（0.28 mg/mL）和乙二醇（8.75 mg/mL）。也可以5 mg/mL溶于DMSO，易溶于DMF和甲酰胺。不建议对制霉菌素溶液进行高压灭菌或无菌过滤。制霉菌素混悬液也有效，37°C下可稳定保存3天。制备在水中的 50 mg/mL 悬浮液，并储存于 -20°C。
