脂多糖(LPS)是革兰氏阴性菌细胞壁的特征组分。LPS及其脂质A部分可通过能够识别常见的病原体相关分子模式(PAMP)的Toll样受体蛋白家族成员Toll样受体4(TLR4)刺激先天性免疫系统细胞。
Biochem/physiol Actions
脂多糖(LPS)分布在细胞膜的外层,并且存在于无包膜菌株的细胞表面。LPS可维持外膜的完整性。此外,还可保护细胞免受胆盐应激和亲脂抗生素的影响。
Preparation Note
产物可溶于水(5mg/ml)或细胞培养基(1mg/ml),产生浑浊的、淡黄色溶液。在经过涡旋和加温至 70-80oC 后,在水溶的盐水中得到更浓缩、但仍然浑浊的溶液(20mg/ml)。脂多糖是在各种溶剂中形成胶束的分子。在水和磷酸盐缓冲盐水中观察到浑浊的溶液。有机溶剂不能提供更澄清的溶液。甲醇产生具有漂浮物的混浊悬浮液,而水产生均匀混浊的溶液。
