免疫原

合成肽链对应小鼠GAPDH氨基酸,通过N端半胱氨酸残基和KLH共轭。对应序列在大鼠中相同,在人中有两个氨基酸的差异。

应用

兔抗GAPDH抗体适合作为使用以下材进行蛋白印迹的一抗:
  • 人癌细胞提取物
  • 少肌症老年小鼠肌肉组织提取物
  • 骨小梁或小鼠心肌样本
  • 小鼠胚胎成纤维细胞
它适用于使用小鼠NIH3T3细胞裂解液进行的免疫沉淀(5-10μg)和使用大鼠NRK细胞进行的间接免疫荧光(工作浓度为5-10μg/mL)。它还适用于使用人HeLa细胞全提取物进行的蛋白印迹(工作浓度为0.1-0.2μg/mL)。

生化/生理作用

酶甘油醛-3-磷酸脱氢酶催化在无机磷酸和NAD存在下,甘油醛磷酸可逆氧化磷酸化,在糖代谢中是主要的能量产生步骤。它可与几种蛋白质结合,包括肌动蛋白、微管蛋白、淀粉样蛋白前体、聚谷氨酰胺肽、DRPLA(齿状核红核苍白球丘脑下部核萎缩)和亨廷顿蛋白。GAPDH形成OCA-S的一部分,其中OCA-S是多组分OCT1(八聚体-基序-结合因子)共激活因子复合物,其可参与S期依赖性组蛋白H2B的转录。该关联可将H2B转录机制与细胞周期调节和细胞代谢状态联系起来。为了应对氧化压力,GAPDH诱发细胞凋亡。

储存及稳定性

想要持续使用,储存在2-8℃,可以最多放1个月。
想要延长储存,工作分装冷冻。不建议反复冻融或存放在无霜冰柜中。如果长期储存时出现轻微浑浊,请在使用前通过离心澄清溶液。工作稀释样本如果在12小时内没有使用,应该丢弃。
