应用

FITC-脂多糖(FITC-LPS)可用于研究LPS跨组织屏障(如结肠上皮细胞)的运动/转运和细胞结合以及脂多糖与荧光测定系统的内化等过程。它可用于评估、鉴定和定量脂多糖(LPS)去除试剂和流程。FITC-LPS偶联物可用于检测LPA结合蛋白和基序。
脂多糖(LPS)是革兰氏阴性菌细胞壁的特征组分。LPS及其脂质A部分可通过能够识别常见的病原体相关分子模式(PAMP)的Toll样受体蛋白家族成员Toll样受体4(TLR4)刺激先天性免疫系统细胞。

生化/生理作用

脂多糖(LPS)位于膜的外层,并且暴露在无包膜菌株的细胞表面。它们有助于维持外层膜的完整性,并保护细胞免受胆汁盐和亲脂性抗生素的破坏。

制备说明

本品可溶于水(5 mg/ml),得到浑浊的黄色至橙色溶液。脂多糖分子可在各种溶剂中形成胶束。在水和磷酸盐缓冲盐水中,可观察到混浊的溶液。使用有机溶剂不会得到更澄清的溶液。以甲醇为溶剂将得到含漂浮物的混浊悬浮液,而以水为溶剂将得到均匀的混浊溶液。 用于细胞培养时,应通过向小瓶(1mg)中加入1ml无菌平衡盐溶液或细胞培养基重悬LPS,并轻轻旋转直至粉末溶解。可通过另外添加无菌平衡盐溶液或细胞培养基,将溶液进一步稀释至所需的工作浓度。
