该酶还与其他蛋白酶一起被用于分解人类肿瘤、小鼠肾脏、人脑和肺上皮组织。它还在在肝、肾灌注研究、胰腺消化、非实质大鼠肝细胞和肝细胞分离等方面有应用价值。Sigma的该酶用于牛小梁和皮质骨I型胶原的消化。[1]它也被用作B组链球菌细胞FALGPA检测的阳性对照。[2]

适用于制备用于测序的单细胞悬液。

生化/生理作用

从组织中有效释放细胞需要胶原酶和中性蛋白酶的共同作用。每摩尔胶原酶由4g原子钙(Ca2+) 酶激活。培养滤液被认为含有分子量在68-130 kDa之间的至少7种不同的蛋白酶。最佳pH范围为6.3-8.8。这种酶通常用于消化组织样本中的结缔组织成分，以释放单个细胞。胶原酶治疗会导致一些细胞死亡。通常，消化浓度从0.1到5 mg/mL不等。为产生令人满意的细胞解离效率并不会造成太多的细胞死亡，其反应时间从15分钟到几个小时不等。最好使用含有钙和BSA的Krebs Ringer缓冲液，Zn2+ 是活性必需的。该酶识别序列 -R-Pro-8-X-Gly-Pro-R-，其中X通常是中性氨基酸。乙二醇双(β-氨基乙基醚)-N,N,N′,N′-四乙酸(EGTA)4;β-巯基乙醇;谷胱甘肽(还原型));巯基乙酸钠;2、2′-联吡啶;8-羟基喹啉、半胱氨酸具有抑制该酶活性的作用。
每摩尔胶原蛋白酶被4g原子钙活化。 它被乙二醇-双（β-氨基乙基醚）-N，NN′,N′-四乙酸，β-巯基乙醇、谷胱甘肽、巯基乙酸和8-羟基喹啉抑制。

注意

所提供产品可在-20℃稳定保存一年。在37℃、50℃和-20℃保存30天，无明显的FALGPA和蛋白酶活性损失。如果快速冷冻分装液(10 mg/mL)并保存于-20℃，胶原蛋白酶原液活性稳定。反复冻融会破坏溶液。 本品在冰上置放7小时以上可保留100%活性。

单位定义

一个胶原蛋白消化单元（CDU）从牛跟腱胶原蛋白中释放的肽链量和1.0 μ亮氨酸，5小时，pH7.4,37℃，在钙离子存在下的茚三酮颜色相同。一个 FALGPA水解单元每分钟，25℃，水解1.0 μmole 呋喃基丙烯酰-Leu-Gly-Pro-Ala。一个中性蛋白酶单元水解酪蛋白，产生颜色和1.0 μmole 酪氨酸每5小时，pH7.5,37℃下产生的相同。一个梭菌蛋白酶单元在DTT存在下，pH7.6,25℃，每分钟水解1.0 μmole BAEE。

制备说明

该产品相当于Mandl,I.,等报道的前40%硫酸铵馏分。（Mandl, I., et al., J. Clin.Invest., 32, 1323 (1953).储备溶液制备可在37°C下，将1-2 mg/mL胶原酶溶解于含有TESCA缓冲液（含50mM TES，0.36mM氯化钙，pH 7.4）.
