放线菌素 D 是一种抑制细胞增殖的抗肿瘤抗生素，具有广泛的应用，包括作为细胞培养中的选择剂，用在抑制 HIV 复制和 PC12 细胞程序性死亡的研究中。 细胞培养应用的使用浓度推荐为 1μg/mL。

生化/生理作用

放线菌素 D 通过与双链 DNA 形成稳定的复合物，抑制 DNA 引发的 RNA 合成并引起 DNA 的单链断裂来抑制细胞的增殖。 研究表明，它是逆转录酶中负链转移步骤的抑制剂。

注意

该产品具有吸湿性和光敏性。 如果在 2-8°C 下避光保存，经 HPLC 测试，可保持至少 15 个月。 未使用的放线菌素 D 稀溶液对光极为敏感，容易吸附到塑料和玻璃上，应丢弃。 但是，浓缩原液的冷冻等分试样至少可以稳定一个月。

制备说明

放线菌素 D 以红色、有光泽的晶体形式出售，可溶于乙腈或丙酮，浓度为 10 mg/mL，溶于 DMSO 中最低浓度为 1 mg/mL。 一些参考文献还显示出在水中的微弱溶解度为 0.5 mg/mL。

    Stock solutions of this product may be prepared at 10 mg/mL in either ethanol (EtOH) or dimethyl sulfoxide (DMSO).

    Stock solutions of this product in DMSO or ethanol should be stored in foil-wrapped vials at -20 °C. In DMSO, solution can be stored for up to 1 month at -20 °C. Do not store in glass vials.

    For cell culture applications, actinomycin D is used as a selection agent and is used in banding techniques to differentiate between different regions of chromosomes. Suggested working concentrations is 1 μg/mL.
