Application

Achromopeptidase from bacteria may be used:

in the homogenization step for genomic DNA extraction from microbial biofilm
in the lysis of gram-positive strains(
for DNA extraction from Nocardia strains

Achromopeptidase is useful for lysis of Gram-positive bacteria that are resistant to lysozyme. Achromopeptidase, from Sigma, has been used for the isolation and molecular characterization of skin and soft-tissue bacterial infections. Product A3422 is from bacteria and is a partially purified powder containing lactose.

Biochem/physiol Actions

Achromopeptidase is a bacteriolytic enzyme and belongs to serine protease family. It is effective for the lysis of gram positive bacteria compared to lysozyme. Achromopeptidase activity is inhibited in the presence of heavy metals. Achromopeptidase based inhibition assay may be useful for screening the presence of heavy metals.

Achromopeptidase is a lysyl endopeptidase that has a molecular weight of approximately 27 kDa. It works best between pH 8.5 -9. Approximately 500-1,500 μn/ml achromopetidase can be used to lyse cells at a density of OD₆₀₀= 0.6 over 2 hours at 37 °C.

Other Notes

One unit will lyse 0.4 μg of Micrococcus lysodeikticus per minute by turbidimetric detection at 600 nm when suspended in buffer at pH 8.0 at 37 °C.

Enzymatic Assay of Achromopeptidase

To measure achromopeptidase activity, this procedure uses bacterial cells and a turbidimetric rate assay. Turbidity is measured at 600 nm using a spectrophotometer.
