M1生物测定

1.使用体外半固体琼脂培养物进行M1生物测定,该培养物在1 mL体积的DME(含20%FCS的0.3%琼脂)中含有约100个细胞。

2.加入100 μL样品或mLIF(等渗盐水中5%FCS中的10(E4)单位/mL),一式两份两倍系列稀释至35 mm培养皿。

3.将100μ L 5%FCS的等渗盐水加入到两个对照载玻片上。

4.在37°C、含10%CO2的完全湿润空气中培养7天。

5.对显示分化的菌落数进行评分(注:50单位定义为导致50%菌落分化的活性量)。
