应用

对于大多数体外应用,bFGF在0.1至10.0 ng/mL的浓度范围内发挥其生物学活性。 响应细胞包括(部分列表): 内皮细胞,间充质细胞。根据培养的方法,人ES细胞所需的浓度在4到100 ng/ml的范围内

外形

从5mM Tris溶液(pH 7.6)和150mM NaCl溶液中冻干。

储存及稳定性

将冻干物料保持在-20°C,直至标签上注明的失效日期。

一般应用:
快速旋转后,在0.1 M磷酸盐缓冲液(pH值6.8)中复溶至0.1-1.0 mg/mL的浓度。 复溶的bFGF应以工作等分试样保存于-20°C中,可保存长达六个月。 多次冷冻/解冻循环会导致大量活性损失。

对于人ES细胞培养:
快速旋转后,在含有0.5%BSA,1 mM DTT和10%甘油的Dulbecco′s PBS过滤溶液中复溶至10 μg/mL。分装并在-20°C下储存长达6个月。 然后,该溶液可以解冻并稀释至4 ng/mL,用于人ES细胞与饲养层的培养,或稀释至8 ng/mL,以补充小鼠胚胎成纤维细胞调节培养基(用于无饲养层的人ES细胞培养)。

分析说明

比活度:与标准品相比,人FGF-b具有完全的生物活性。 ED50是通过表达FGF受体的NIH3T3细胞对胸苷摄取的剂量依赖性刺激来确定的。
