General description
II型胶原蛋白是一种纤维状胶原蛋白，它是透明软骨的主要成分，占胶原蛋白的95%和软骨干重的60%左右。它是一种同三聚体，由位于人类染色体12q13的COL2A1基因编码。2型胶原蛋白也主要存在于椎间盘髓核和眼睛的视网膜、巩膜和水晶体中。IIA型主要位于眼睛的玻璃体中。IIB型优先在成人软骨组织中产生。 采用差盐沉淀法、醇沉淀法和DEAE色谱法纯化II型胶原蛋白。多重纯化步骤得到了天然三螺旋形态的胶原蛋白类型的最终制剂，不含任何非胶原蛋白。其他类型的胶原蛋白在标准制剂中的污染要低于百分之五。
Application
人II 型胶原蛋白已用于：
- 作为微阵列分析中的细胞外基质（ECM）蛋白，以包被硝化纤维膜载玻片进行探测和检测实验
- 研究Stickler综合征斑马鱼模型中Col2a1a 在眼早期发育神经嵴中的作用
- 作为校准标准品用于通过酶联免疫吸附试验(ELISA) 定量测定骨架中的 II 型胶原蛋白水平
Biochem/physiol Actions
II型胶原蛋白通常与XI胶原蛋白共组装，形成一种与IX胶原蛋白共价交联胡复合物，并与富含亮氨酸的小分子蛋白聚糖相互作用。2型胶原蛋白赋予组织稳定性和强度，并维持压力下的结构完整性和弹性。II型胶原蛋白主要通过基质金属蛋白酶胶原酶的作用裂解，导致生成众所周知的生物标志物，如II型胶原蛋白的C末端端肽和C2C。II型胶原蛋白的变异导致软骨发育异常，从而引起早发性骨关节炎。
