胱天蛋白酶-3（CPP32，Apopain）是研究最广泛的凋亡蛋白。胱天蛋白酶-3被合成为一种无活性的酶原（32 kDa），该酶在经历凋亡的细胞中通过自身蛋白水解而处理和/或被另一种上游蛋白酶切割而处理。胱天蛋白酶-3的加工形式由大（17 kDa）和小（12 kDa）亚基组成，它们结合形成活性酶。活性胱天蛋白酶-3蛋白水解切割并激活其他胱天蛋白酶，以及细胞中的相关靶标，例如PARP和DFF。

蛋白质印迹分析：
我们用活性凋亡细胞的1：100-1：200（0.5-4µg/ml）稀释液来检测胱天蛋白酶-3。
注意当使用组织时，活性胱天蛋白酶3的水平可以低于可检测的水平。使用CC119（人胱天蛋白酶3）与1 mM ATP培养30分钟（用于激活），作为westerns上的阳性对照。或者，用星形孢菌素（0.5µM，10-18小时）培养的神经元培养制剂通常会产生很强的胱天蛋白酶3信号。

免疫组化：
1：10稀释度（10-20µg/ml），AB3623在4%PFA固定、石蜡包埋的小鼠组织中反应成功。

免疫组化（石蜡）：
胱天蛋白酶-3单克隆抗体的最佳染色： 小鼠小肠

生化/生理作用

AB3623仅检测裂解的p17片段，而不检测经历凋亡的细胞中的前体形式。

在含有PBS，30%甘油，0.5%BSA，5 mM EDTA和0.03%proclin的缓冲液中纯化的兔多克隆抗体。
